They then extend their axons to the median eminence (ME) to allow secretion of GnRH into the hypophyseal portal system. From the OP, they migrate across the cribriform plate and through the basal forebrain to arrive within the presumptive hypothalamus by E17.5. GnRH neurons have a unique point of origin within the olfactory placode (OP) at embryonic day 11.5 (E11.5) ( Schwanzel-Fukuda and Pfaff, 1989 Wray et al., 1989 Schwanzel-Fukuda et al., 1992). GnRH stimulates the anterior pituitary gland to secrete gonadotropins, which, in turn, act on the gonads to control gametogenesis. Mammalian reproduction is mediated by the pulsatile release of gonadotropin-releasing hormone (GnRH) from a distinct population of neurons within the hypothalamus. Together, these data indicate that Si圆 plays an important role in the regulation of GnRH expression and hypothalamic control of fertility. Finally, we demonstrate that this induction in GnRH expression is mediated via binding of Si圆 to evolutionarily conserved ATTA sites located within the GnRH proximal promoter. We show that expression of Si圆 is dramatically increased during GnRH neuronal maturation and that overexpression of Si圆 induces GnRH transcription in neuronal cells. Although basal gonadotropin production in these mice is relatively normal, analysis of GnRH expression reveals a dramatic decrease in total GnRH neuron numbers. Female Si圆-null mice exhibit a striking decrease in fertility, failing to progress through the estrous cycle normally, show any signs of successful ovulation, or produce litters. In this study, we identify sine oculis-related homeobox 6 (Si圆) as a novel factor necessary for proper targeting of GnRH expression to the limited population of GnRH neurons within the adult mouse hypothalamus and demonstrate that it is required for proper reproductive function in both male and female mice. If they aren’t, clean the second piece of glass, then put it down over your negative. ![]() Wait, you are wondering what that second piece of glass was for, right? Well, as we don’t use a carriage for our negatives, chances are they are not totally flat. As you read these lines, I’m writing a second article that will detail this process for you. If you don’t know how to do this, be sure to stay tuned. All you have to do next is convert it to a positive image in Capture One, Lightroom, or Photoshop. You now have you negative in digital form. To do so, switch on the live view mode and zoom in all the way. Make sure to focus on the grain and not on the picture itself as it may have been out of focus in the first place. Once everything is cleaned, you can go ahead and snap a picture of your negative. An air blower will probably come in handy here. Also, make sure both the glass, the negative, and the screen are pristine. Depending on the aperture you are using, you might have to raise the negative even more than just a few millimeters. The glass on top of it is here just to separate the negative from the screen and avoid having the pixels showing in our scan. So make sure to turn its brightness to the maximum, and have a pure-white image displayed in full-screen mode. If you hadn’t understood yet, the tablet is meant to light our negative from underneath. At this stage, your setup should look something like a poor man's reprographic system. To be honest with you, this is the hardest part. The lens should be pointing straight down, and the focal plan must perfectly parallel to the glass surface, where your negative will be placed. Place your tablet (or phone or laptop) on a table, install one of the two glasses on top of the screen, and have your camera on the tripod above it. The initial idea actually came from seeing this video: The goal here is to recreate a reprographic system with a light table. Now that we are clear with what we need let’s set it all up. Also, if you have everything except the macro lens, it will still cost you less than a good Epson or Imacon scanner. Some of you may be lacking the macro lens, but it can be replaced by a 50mm with extension tubes (just be careful with deformation and vignetting then). So while my technique remains very basic, it will cost you less than $2,000 and will yield digital files that are comparable. If you were to buy a second-hand Imacon, it would set you back $2,000. An expensive machine but not the best around either - not as good as a drum scanner. Photos from your grandparents' albums, your parents' wedding, photos of your first birthday, your child’s 1st birthday, Thanksgiving, those are the ones you want to scan and preserve forever. Larger scans take longer so that is factored into the price also. We ask what size you'd like the final print to be, and we scan your original to produce a file large enough to make that final print. It depends on how many originals you have to be scanned, what condition they're in and if any restoration is needed. Pricing is determined on a case by case basis. A 35mm slide measures 24mm by 36mm, to make a 5x7 print, the slide needs to be enlarged 400% so any and all flaws are magnified by 400% also. We can scan your precious slides, but we may need to do extensive clean up work on them. When you squint at your slides in the sun or on a light table, they may look ok, but dust, mold, and fungus LOVE slides. We can scan your old photos, retouch rips and stains if needed, restore the color/tone, and produce new prints for you and copies for the rest of the family. I've played cRPGs for many years now, back to BG1 and possibly before. I got more than enough Might and Speed points in my group for the occasionally dialog based skill check. It just makes the game very easy, almost too easy, to play like this. ![]() In essence, despite the sub-optimal build and having the combat prowess of a wet noodle, I do just fine. I don't need to collect gear, I don't really need to understand the particularities of combat skills and I keep Aligern and two others around for skill checks and conversation. I don't always get what I want out of conversations (I'll get back to that as well), but manage to do pretty well most of the time. She can basically talk her way out of a war crimes tribunal if she wanted to and I haven't once had to use any cyphers or other boosters to get past a skill check. Regarding the build(s), for the main character I did try to focus on intelligence, conversational skills and cyphers that also emphasize these traits. The few times I've experimentally tried starting a fight, I get utterly and totally creamed. Most fights can be talked around entirely without starting a crisis. I haven't managed to avoid being hit inside the crises entirely due to a sub-optimal build (I'll get back to that) but a combination of running, hiding and having just enough hitpoints have made getting through pretty easy. ![]() I'm playing a charming jack with a silver tongue, something like that, and I've managed to cruise through without any actual combat except twice in optional quests (one of which Nekojin and I discussed above). So you might have a 70/30 weave or 60/40 weave or no, still 50/50 depending upon the sail but we changed the way that the cloth is used with the Dacron polyester fibre. Today, we really mix the weave construction mix. It used to be, when Ted Hood pioneered this back in the late 50’s, it was a cross cut, XY axis type of weaving and the same fibre was on the X axis as was on the Y axis. It’s a lot of different weights, lots of different styles, and lots of different weaves. We’ve used it forever, there had been lots of advances and it’s been a more engineered product in the last 10, 15 years than it has been. Within the cloth and we’ll have lots of different types of cloth but the basic fibres we use, and the fibre is what’s going to dictate how strong the cloth is, we’re going to have Dacron or polyester, this has been tried and true. With that in mind, I’m going like try and think about, what does it take, what goes in to making a good sail? What do we have to think about? The foundation of any good sail is going to start with sail fibre, the cloth. So it really makes a difference in how we approach the sail we’re going to build. So all these things factor into, as a sail maker, things that I consider when I build an offshore sail versus when I build a sail for someone who is bay cruising or even sometimes coastal cruising. ![]() The sails get a lot more demands on them a lot of wear and tear, a lot of weathering, a lot more chafe, even just the acts of putting a reef in has a lot of wear and tear, takes a lot of life out of sails. You don’t get to choose what sea state or what wave, or what wind conditions you sail in. You’re making a long passage, all of a sudden your sails shift from being used in good conditions or fair conditions to now you’re going to have periods of extended times in UV extended wear and tear. Your choices or things are dictated to you more. When you go off shore, your bringing all these in here. With that, offshore sailing, what does it mean to me as a sail maker? As I mentioned, it’s a very different approach and basically, when you’re on the bay, you’re using your sails but you get to pick and choose when you go out, you get to pick and choose the conditions you sail in, it’s very different. But we’re going to try and really address offshore sails and what you need to think about, what you need to start preparing for and how using sails offshore is very different from maybe how you’ve been using them currently.
